ABSTRACT

Abstract The podcast Femigrantes BR is defined as "a space for conversation among feminist-immigrant women around the world". The podcast had 21 episodes, gathering stories of Brazilian women who live and work in several countries. In the voices of the interviewees, life stories expand beyond the personal account and connect with firmly established structures such as racism and the unequal relations between the Global North and South. Taking that object and scenario, I investigate what has been the contribution of feminisms, as political discourse, to the "empowerment" of those migrant women and analyse how the media coverage of the feminist debate reflects on the way they act in the public life of the country in which they now live. In the effort to attend to these points, I engage in an immersion in digital media and, in dialogue with contributions from feminist media, technology, decolonial, postcolonial studies, I discuss how the feminist occupation of the internet can potentiate social transformations in unequal gender structures and contribute to challenge hierarchies between North and Global South.

ABSTRACT

OBJECTIVE:To study the effects of costunolide (COS)on the cell cycle distribution and apoptosis of human breast cancer SK-BR- 3 cells and its mechanism. METHODS :Human breast cancer SK-BR- 3 cells were divided into blank control group, and COS groups of 10,20,30,40,50 μmol/L. MTT assay was used to detect the effects of COS on cell proliferation. SK-BR-3 cells were divided into blank control group ,COS low ,medium and high concentration groups (10,20,30 μmol/L). After cultured for 24 h,flow cytometry was used to detect the distribution of cell cycle. Hoechst 33258 fluorescence staining was used to detect cell apoptosis. Western blot assay was used to detect the expression of p 53,caspase-3,Bcl-2,Bax,p21,CDK2 and cyclinE. RESULTS :Compared with blank control group ,COS could significantly inhibit the proliferation of SK-BR- 3 cells(P< 0.05 or P<0.01),and in a dose and time-dependent manner. Low ,medium and high concentrations of COS could induce cell apoptosis and arrest cell at G 1/S phase (P<0.05 or P<0.01),could significantly up-regulate the protein expression of p 53, caspase-3,Bax and p 21(P<0.05 or P<0.01),and could significantly down-regulate the protein expression of Bcl- 2,CDK2 and cyclinE(P<0.01). CONCLUSIONS :COS can inhibit the proliferation of human breast cancer SK-BR- 3 cells and induce cell apoptosis and cell cycle arrest. The mechanism may be related to the regulation of p 53/Bax/Bcl-2/caspase-3 apoptosis signal pathway.

ABSTRACT

Breast cancer brain metastases (BCBMs) are one of the most difficult malignancies to treat due to the intracranial location and multifocal growth. Chemotherapy and molecular targeted therapy are extremely ineffective for BCBMs due to the inept brain accumulation because of the formidable blood‒brain barrier (BBB). Accumulation studies prove that low density lipoprotein receptor-related protein 1 (LRP1) is promising target for BBB transcytosis. However, as the primary clearance receptor for amyloid beta and tissue plasminogen activator, LRP1 at abluminal side of BBB can clear LRP1-targeting therapeutics. Matrix metalloproteinase-1 (MMP1) is highly enriched in metastatic niche to promote growth of BCBMs. Herein, it is reported that nanoparticles (NPs-K-s-A) tethered with MMP1-sensitive fusion peptide containing HER2-targeting K and LRP1-targeting angiopep-2 (A), can surmount the BBB and escape LRP1-mediated clearance in metastatic niche. NPs-K-s-A revealed infinitely superior brain accumulation to angiopep-2-decorated NPs-A in BCBMs bearing mice, while comparable brain accumulation in normal mice. The delivered doxorubicin and lapatinib synergistically inhibit BCBMs growth and prolongs survival of mice bearing BCBMs. Due to the efficient BBB penetration, special and remarkable clearance escape, and facilitated therapeutic outcome, the fusion peptide-based drug delivery strategy may serve as a potential approach for clinical management of BCBMs.

ABSTRACT

Objective:To evaluate the effect of down-regulating lncRNA LINC00263 targeting miR-4458 on the proliferation, migration, invasion and radiosensitivity of breast cancer SK-BR-3 cells. Methods:The expression differences of LINC00263 in breast cancer tissues, adjacent tissues, normal breast epithelial cells and breast cancer cells were determined by qRT-PCR. Transfection of LINC00263 shRNA in breast cancer SK-BR-3 cells down-regulated the expression of LINC00263, and the cloning experiment was used to detect the radiosensitivity. Breast cancer SK-BR-3 cells were treated with 6 Gy irradiation. CCK-8 assay was employed to detect cell proliferation. Flow cytometry was adopted to detect cell apoptosis. Transwell chamber test was performed to detect cell migration and invasion. Western blot was used to detect the expression levels of C-Caspase-3 and C-Caspase-9, MMP-2 and MMP-9 proteins. Bioinformatics software predicted that LINC00263 and miR-4458 had complementary binding sites, and the luciferase reporter system was utilized determine the targeting relationship between LINC00263 and miR-4458. LINC00263 shRNA and miR-4458 inhibitor were co-transfected into breast cancer SK-BR-3 cells, and 6 Gy irradiation was given to detect the changes in cell proliferation, apoptosis, invasion and migration. Results:The expression level of LINC00263 in breast cancer tissues was higher than that in adjacent tissues. The expression level of LINC00263 in breast cancer cells was higher compared with that in normal breast epithelial cells. The radiosensitivity of breast cancer SK-BR-3 cells was increased after transfection of LINC00263 shRNA. Transfection of LINC00263 shRNA and radiation exerted a synergistic effect, jointly inhibited breast cancer cell proliferation, migration and invasion, promoted cell apoptosis, up-regulated the expression levels of C-Caspase-3 and C-Caspase-9 proteins in cells, and down-regulated those of MMP-2 and MMP-9 proteins. Down-regulation of LINC00263 targetedly up-regulated miR-4458 expression. miR-4458 inhibitor reversed the inhibitory effect of LINC00263 shRNA combined with radiation on the proliferation, migration, invasion and apoptosis promotion of breast cancer SK-BR-3 cells.Conclusion:Down-regulating lncRNA LINC00263 targeting miR-4458 inhibits the proliferation, migration and invasion of breast cancer SK-BR-3 cells, and improves cell radiosensitivity.

ABSTRACT

The objective was to compare the dry matter consumption (CMS) observed, through the use of the GrowSafe® electronic system, with that predicted by the BR-Corte (2010 and 2016) and NRC (2000) nutritional systems in confined Senepol bulls. To this end, 24 Senepol Bulls were used in a completely randomized design, uncastrated with an average initial weight of 368 kg and 16 months of age. The evaluation of the accuracy and approximation of the CMS estimates by the nutritional systems was adjusted by the simple linear regression model and the decomposition of the mean square of the prediction error (QMEP). The mean CMS observed was 10.33 kg.day-1, higher than the values predicted by the nutritional systems, in which the values predicted by the NRC (2000) and BR-Corte 2010 and 2016 underestimated the CMS by 29.62, 6.19 and 2.03%, respectively. The verification of QMEP and its decomposition made it possible to infer the proximity of the values predicted by the BR-Corte 2010 and 2016 models and the values observed, which presented a better adjustment in relation to the NRC. Surprisingly the values predicted by the NRC, created from a database with Bos taurus animals, showed greater distance from the values predicted and observed, and it was expected greater accuracy of the NRC models for this category and animal breed. It is concluded that the BR-Corte 2016 was the most appropriate model to estimate the CMS of confined Senepol bulls.

ABSTRACT

Breast cancer is the most common malignancy in females and an important cause of cancer deaths worldwide. With advances in oncopathology, breast cancers are now diagnosed and treated at a molecular level. To distinguish and subdivide the type of cancer with the suitable markers and to foresee its prognosis and therapeutic approach, cytokeratin can be used which is prognostic tumour marker and has a number of different advantages as it is an important IHC marker, cytokeratin (Ck) is epithelial intermediate filament, which is expressed in a tissue specific manner. Expression of the intermediate filaments, predominantly cytokeratins (Ck) reflects the epithelial cell type. In breast, the luminal epithelial cells express CK 8 / 18, CK 7 and CK 19, while basal cell expresses CK 5 / 6, CK 14 and CK 17. Bloom Richardson grading (BR) system has a powerful prognostic value. It is also incorporated in the algorithm for the determination in the use of adjuvant chemotherapy. Moreover, as a tumour marker cytokeratin can accurately predict disease status as it is easy, inexpensive and a reliable tool for efficient management. CK and BR grading can be used as cost effective diagnostic tools in hormone positive breast carcinoma for diagnosis as well as treatment. Very early detection of breast cancer reduces the mortality and morbidity.

ABSTRACT

OBJECTIVE:To study the effects of costunolide on the proliferation ,migration and apoptosis of breast cancer SK-BR-3 cells and its mechanism. METHODS :SK-BR-3 cells in logarithmic growth period were collected and cultured with different concentrations (10,20,30,40,50 μmol/L)of costunolide for 24,48,72 h. Inhibitory rate of costunolide on cell proliferation was detected with CCK- 8. The cells were divided into blank control group and costunolide group (10,20,30 μmol/L). Hoechst 33258 fluorescence was used to observe the morphology and apoptosis of cells ,and apoptotic rate of cells were calculated. Cell scratch test was used to detect the migration ability of cells and calculate the migration rate. Western blotting was used to detect the relative expression level of Bcl- 2,Bax,Caspase-3 and Cleaved Caspase- 3 in cells. RESULTS :The proliferation of SK-BR-3 cells were significantly inhibited by costunolide (P<0.05 or P<0.01),and it shows a trend of concentration and time dependence. In the blank control group ,cells possessed clear contour ,regular shape and good adherence . Compared with blank control group, the number of cells were decreased significantly in 10,20,30 μmol/L costunolide groups, the cell structure was loose, the volume was reduced ,and the gap became larger ,and most of the cell contour disappeared and became round ,the cell adherence was poor ;cell migration rate and Bcl- 2 protein relative expression level were decreased significantly ,while apoptosis rate and the relative expression level of Bax ,Caspase-3 and Cleaved Caspase- 3 protein were significantly increased (P<0.05 or P<0.01). CONCLUSIONS : Costunolide can inhibit the proliferation and migration ,and induce apoptosis of human breast cancer SK-BR- 3 cells, mechanism of which may be through up-regulating the expression of Bax ,Caspase-3 and Cleaved Caspase- 3 while down-regulating the expression ofBcl-2.

ABSTRACT

ABSTRACT. The Movement Disorder Society has published some recommendations for dementia diagnosis in Parkinson disease (PD), proposing the Montreal Cognitive Assessment (MOCA) as a cognitive screening tool in these patients. However, few studies have been conducted assessing the Portuguese version of this test in Brazil (MOCA-BR). Objective: the aim of the present study was to define the cut-off points of the MOCA-BR scale for diagnosing Mild Cognitive Impairment (PD-MCI) and Dementia (PD-D) in patients with PD. Methods: this was a cross-sectional, analytic field study based on a quantitative approach. Patients were selected after a consecutive assessment by a neurologist, after an extensive cognitive evaluation, and were classified as having normal cognition (PD-N), PD-MCI or PD-D. The MOCA-BR was then applied and 89 patients selected. Results: on the cognitive assessment, 30.3% were PD-N, 41.6% PD-MCI and 28.1% PD-D. The cut-off score on the MOCA-Br to distinguish PD-N from PD-D was 22.50 (95% CI 0.748-0.943) for sensitivity of 85.5% and specificity of 71.1%. The cut-off for distinguishing PD-D from MCI was 17.50 (95% CI 0.758-0.951) for sensitivity of 81.6% and specificity of 76%.

ABSTRACT

It is evident that sedentary behavior is related to the occurrence of several diseases and increased risk of death. Specifically in the young population, studies show that the levels of physical activity and physical fitness of children and adolescents are decreasing. Information about the performance and quality of the motor actions in children and adolescents are necessary to physical education, sports and general physical activities programs elaboration. Therefore, there are different tools of evaluation that aim to observing the motor behavior characteristic, among then, the Function Movement Screen and the Projeto Esporte Brasil battery of tests. This study aimed to verifying the association between Functional Movement Screen score and the physical fitness variables of the Projeto Esporte Brasil battery of tests in schoolchildren. Participants were 24 schoolchildren (15 boys) of 12 years old. Data were described using mean, standard deviation (parametric variables), median and interquartile difference (non-parametric variables). For the association between Functional Movement Screen score and physical fitness variables were used Pearson's coefficient correlations (parametric variables) and Sperman's coefficient correlation (non-parametric variables). Significant and moderate associations were found between Functional Movement Screen score and flexibility (r=.69; p=.004), lower limb power (r=.68; =.005) and sprint test (r=-.57; p=.025) only in boys. These results evidence a limited capacity of the Functional Movement Screen to be associated to physical fitness in schoolchildren...(AU)

ABSTRACT

With the arrival of new early genotypes and high nutritive value and productive potential, millet has been gaining prominence in recent years for the production of silage, and it promises lower costs of production. Thus, the consortium of millet with tropical forages can increase the production of silage, providing more food to be used in the offseason, where there is low availability of forage. The objective of this study was to evaluate the dry mass production, fermentative characteristics and chemical-bromatological of pearl millet silage and Paiaguas palisadegrass in monocropping and intercropping in different forage systems in the second cropping season. The experimental design consisted of randomized blocks with four replicates. The treatments consisted of silage of the following forage systems: monocropped pearl millet, monocropped Paiaguas palisadegrass, pearl millet intercropped in rows with Paiaguas palisadegrass, pearl millet intercropped inter-row with Paiaguas palisadegrass, and pearl millet oversown and intercropped with Paiaguas palisadegrass (totaling 20 experimental plots). The consortium of pearl millet with Paiaguas palisadegrass contributed to raise the dry matter contents of the silages, except in the system using the overgrowth. Pearl millet silages monocropped and intercropped with Paiaguas palisadegrass presented reductions in pH and ammoniacal nitrogen values as well as favor lactic fermentation. Pearl millet monocropped and intercropped silage exhibited better fermentative and bromatological characteristics than silage obtained from monocropped Paiaguas palisadegrass. Therefore, creating silage from intercropped forages provides an interesting supplemental roughage option that can be used during the offseason for animal feeding.

ABSTRACT

Objective To establish a quantitative analysis of multi-components with single marker (QAMS) for the simultaneous determination of chlorogenic acid, cryptochlorogenin acid, caffeic acid, hyperoside, isoquercitrin, astragalin, kaempferol in crude and processed Cuscuta australis, which is proved to be a scientific and feasible method in the quality analysis in C. australis. Methods Six relative correction factors (RCFs) of chlorogenic acid, cryptochlorogenin acid, caffeic acid, isoquercitrin, astragalin, kaempferol was established in the HPLC method with the hyperoside as the internal standard (IS), which was to calculate the mass fraction of each. The mass fraction of seven effective constituents in crude and processed C. australis was calculated by the external standard method (ESM) at the same time. Compared with the content results determined by the ESM and QAMS, the feasibility and accuracy of QAMS method were verified. Results The relative correction factor (RCF) was perfect. The detection calculated by QAMS was consistent with the results by ESM. Conclusion The method with a single marker, using the hyperoside as IS, is accurate and feasible for the quantitative analysis of six other effective constituents in C. australis.

ABSTRACT

Objective To establish the fingerprint profiling of fruits of Macleaya cordata and study the method for its quality evaluation. Methods The fingerprint profiling of M. cordata fruits from different regions was established using high performance liquid chromatography (HPLC) based on the local standard which was the determination method of the content of protopine, allocryptopine, sanguinarine, and chelerythrine from Changsha of Hunan Province in 2009. The principal component analysis (PCA) and cluster analysis (CA) were performed to explore the correlation among the common fingerprint peaks, origins, and quality of M. cordata fruits. Results Eleven common fingerprint peaks were identified in the fingerprint profiling of chemical constituents of M. cordata fruits from different regions. M. cordata fruits produced from eight areas were classified into two classes by PCA and CA method, and there were five common peaks, including peak 5, 7, 8, 9, and 11 with significant contribution on the regional difference of the fingerprint. Also, common peak 6 was the right peak as the reference peak because of its less variation, appropriate retention time and intensity. Conclusion The fingerprint profiling of chemical constituents of M. cordata fruits established in this study has good precision, repeatability, and stability, which can be used to evaluate the quality of fruits of M. cordata from different producing areas.
